Gut says the first is right and the second nonsensical but, of course, one's gut can make some odd choices and I just noticed myself undetmining my argument in this sentence.

I'd say they have slightly different meanings.

"Of course you're lying" - subtext - because you're a lying shit, and you always lie.

"Of course, you're lying" - subtext - realisation dawns, God, I've been an idiot, you're not telling the truth.
